Abstract | The structural characterization of O-linked glycans present on bacterial flagellins was analyzed. The modified peptides were identified by combination of enzymatic cleavage and LC/MS analysis. The flagellins were digested by trypsin which were analyzed using liquid chromatography coupled to nanoelectrospray mass spectrometry on Q-TOF instrument. The nanoLC/MS-MS experiment provided peptide sequence coverage map of 96% and glycosylation site was identified using alkaline hydrolysis. |
