| Abstract | Hybridization of somatic cells has been suggested as a possible technique to overcome the sexual incompatibility of two unrelated plant species. Somatic hybrids might be of great value for crop improvement (Schenk and Hildebrandt, 1968). Hybridization of somatic plant cells by means of protoplast fusion requires the following steps: (a) removal of the cell wall to form protoplasts; (b) high frequency of protoplast fusion; (c) identification and selection of hybrids; (d) culture of the hybrid protoplasts; (e) regeneration of plants from the hybrid protoplasts.
Attempts to hybridize somatic cells were made early in this century by Winkler, Kiister, and Michel (in Power eta!., 1970; Melchers and Labib, 1974). Michel (1937) demonstrated the fusion of homo- and heterospecific protoplasts after their treatment with sodium nitrate solution. However, fusions were rare and the subsequent fusion products could not be cultured (Power et a!., 1970).
Rapid advances in techniques of protoplast production, fusion, and culture have been made during the last few years, some of which will be reported here. |
